Appendix Sheets 3.1: Recovery of Decomposed Pig Leg Metabolite Peak IDs using MultiQuant and MS-DIAL software
Metabolites were extracted from both skin and muscle layers of a pig leg over seven timepoints, from initial zero hour (0HR) to of a 42 days (42D), and ran on a SCIEX 6600 TripleTOF Q-TOF mass spectrometer. Data acquired using SWATH-DIA were processed using both commercial (SCIEX’s MulitQuant 3.0.2, ins. ref ) and open-source (MS-DIAL 4.9) software. Peaks from MS1 and MS2 were matched against a metabolite library of 235 standards in MultiQuant and scored using a custom VBA macro (a). MS2 (b) and MS1 (c) peaks were processed independently after MS-DIAL, with sorting and filtering based on identification scores. In all cases, green results show a high confidence of identification, yellow a lower confidence and red indicates missing IDs.
